Progesterone in Oil (PIO) is a medication that consists of the hormone progesterone, which is dissolved in an oil base, typically sesame oil or cottonseed oil. It is commonly used in various medical treatments, especially in assisted reproductive technologies (ART) like in vitro fertilization (IVF).

Key Uses of PIO:

  1. Fertility Treatments:

    • PIO is frequently used in IVF and other ART procedures to support the luteal phase of the menstrual cycle (the second half) after egg retrieval or embryo transfer. Progesterone plays a crucial role in preparing the uterine lining for embryo implantation.

  2. Hormonal Therapy:

    • It may also be used in hormone replacement therapy (HRT) for women undergoing menopause, as part of the management of postmenopausal symptoms, often in combination with estrogen.

Administration:

  • PIO is typically administered through intramuscular (IM) injections, usually in the gluteal muscle (buttocks). The injections are usually given daily or multiple times a week during fertility treatments, depending on the specific protocol prescribed by the doctor.

Common Side Effects:

  • Injection site reactions: Pain, redness, swelling, or irritation at the injection site.

  • Fatigue and headaches.

  • Mood changes: Hormonal changes may affect mood, leading to feelings of irritability or depression.

  • Bloating or breast tenderness.

  • Skin reactions: Sometimes, the oil can cause lumps or hard areas in the muscle where the injection is given.
